Do\u00e7. Dr. Esra \u00c7a\u011favi&#8217;nin y\u00fcr\u00fctt\u00fc\u011f\u00fc \u00e7al\u0131\u015fmada, Nobel \u00f6d\u00fcll\u00fc &#8220;Yeniden Programlama&#8221; tekni\u011fi kullan\u0131larak hasta k\u00f6kenli ve uyar\u0131lm\u0131\u015f k\u00f6k h\u00fccre kaynakl\u0131 kalp kas dokusu \u00fcretiliyor.<\/p>\n<p>Kal\u0131tsal kalp hastal\u0131klar\u0131n\u0131n ara\u015ft\u0131r\u0131lmas\u0131 ve yeni tedavi yakla\u015f\u0131mlar\u0131n\u0131n geli\u015ftirilmesi amac\u0131yla y\u00fcr\u00fct\u00fclen \u00e7al\u0131\u015fma, Medipol \u00dcniversitesi Rejeneratif ve Restoratif T\u0131p Ara\u015ft\u0131rmalar\u0131 Merkezi (REMER) b\u00fcnyesinde T\u00dcB\u0130TAK&#8217;\u0131n deste\u011fiyle ger\u00e7ekle\u015ftiriliyor.<\/p>\n<p>\u00c7al\u0131\u015fmada, hastalardan al\u0131nan kan h\u00fccreleri, embriyonik k\u00f6k h\u00fccrelere d\u00f6n\u00fc\u015ft\u00fcr\u00fclerek yeniden programlan\u0131yor. Bu teknikle geli\u015ftirilen k\u00f6k h\u00fccrelerden, laboratuvar ortam\u0131nda ki\u015fiye \u00f6zel kalp dokusu \u00fcretilerek, hastal\u0131klar\u0131n modellenmesi ve h\u00fccresel tedavi kayna\u011f\u0131 olu\u015fturulmas\u0131 ama\u00e7lan\u0131yor.<\/p>\n<p>Doku sayesinde, kalp hastal\u0131klar\u0131 tedavisinde kullan\u0131labilecek potansiyel metot ve ila\u00e7lar\u0131n, bundan sonra s\u0131n\u0131rs\u0131z elde edilebilen ki\u015fiye \u00f6zel h\u00fccreler vas\u0131tas\u0131yla geli\u015ftirilebilmesi hedefleniyor.<\/p>\n<p>&#8211; &#8220;Kandan elde edilen h\u00fccreler, kalp kas\u0131 h\u00fccresine d\u00f6n\u00fc\u015ft\u00fcr\u00fcl\u00fcyor&#8221;<\/p>\n<p>\u00c7al\u0131\u015fmaya ili\u015fkin AA muhabirinin sorular\u0131n\u0131 yan\u0131tlayan REMER Kurucu M\u00fcd\u00fcr\u00fc Prof. Dr. G\u00fcrkan \u00d6zt\u00fcrk, bir\u00e7ok \u00e7al\u0131\u015fman\u0131n y\u00fcr\u00fct\u00fcld\u00fc\u011f\u00fc merkezin, nitelikli \u00e7ok say\u0131da bilim insan\u0131n\u0131 da b\u00fcnyesinde bar\u0131nd\u0131rd\u0131\u011f\u0131n\u0131 s\u00f6yledi.<\/p>\n<p>\u00d6zt\u00fcrk, merkez b\u00fcnyesinde y\u00fcr\u00fct\u00fclen kalp kas\u0131 dokusu \u00e7al\u0131\u015fmas\u0131n\u0131n da T\u00dcB\u0130TAK&#8217;\u0131n deste\u011fiyle s\u00fcrd\u00fcr\u00fclen \u00f6nemli bir proje oldu\u011funa i\u015faret ederek, \u015funlar\u0131 kaydetti:<\/p>\n<p>&#8220;Bu projede, \u00f6zellikle ritim bozuklu\u011fuyla do\u011fan \u00e7ocuklar\u0131n hastal\u0131klar\u0131na tam te\u015fhis koyabilmek ve en do\u011fru tedaviyi se\u00e7ebilmek i\u00e7in bu hastal\u0131\u011f\u0131 laboratuvar ortam\u0131nda modelleyebilme hedefi var. Bu \u00e7ok zor bir \u015fey. Bug\u00fcn bilim d\u00fcnyas\u0131n\u0131n en \u00fcst teknolojilerinden birisi, insandan ald\u0131\u011f\u0131n\u0131z kan h\u00fccrelerini \u00e7ok zahmetli bir i\u015flemden sonra organa ait h\u00fccrelere d\u00f6n\u00fc\u015ft\u00fcrmektir. Bahsetti\u011fimiz projede de ritim bozuklu\u011fuyla do\u011fan \u00e7ocuklardan al\u0131nan kanlardan elde edilen h\u00fccreler, kalp kas\u0131 h\u00fccresine d\u00f6n\u00fc\u015ft\u00fcr\u00fcl\u00fcyor. Bu h\u00fccreler, k\u00fclt\u00fcr ortam\u0131 dedi\u011fimiz yapay ortamda birbirleriyle irtibatlar\u0131n\u0131 kurup, minyat\u00fcr doku par\u00e7ac\u0131klar\u0131 halinde k\u00fclt\u00fcr kab\u0131nda at\u0131m da g\u00f6stermeye ba\u015fl\u0131yor. Bunlardan biz EKG benzeri kay\u0131tlar alabiliyoruz. Bu h\u00fccrelerdeki ritim bozukluklar\u0131n\u0131 analiz edebiliyoruz. Dahas\u0131 da o hastaya \u00f6zel hangi ilac\u0131n uygun oldu\u011funu test edebiliyoruz. Bunlar\u0131n hepsi hasta kan\u0131 verdikten aylar sonra laboratuvar ortam\u0131nda elde etti\u011fimiz sonu\u00e7lar. Ancak bir \u00e7ocu\u011fu d\u00fc\u015f\u00fcnecek olursan\u0131z, onun t\u00fcm hayat\u0131n\u0131 de\u011fi\u015ftirecek bir s\u00fcreci asl\u0131nda ald\u0131\u011f\u0131m\u0131z bir kanla ba\u015flat\u0131p, \u015fu an uygulanabilecek en ileri teknolojiyle ona faydal\u0131 bir tedavi se\u00e7ene\u011fine kadar g\u00f6t\u00fcrebiliyoruz.&#8221;<\/p>\n<p>\u00c7al\u0131\u015fmayla, tedavi s\u0131ras\u0131nda mevcut ila\u00e7lardan en uygununu se\u00e7ebilme olana\u011f\u0131 yakaland\u0131\u011f\u0131n\u0131 vurgulayan \u00d6zt\u00fcrk, &#8220;\u015eu an bu projede elde etti\u011fimiz ba\u015far\u0131 olduk\u00e7a y\u00fcksek. Projenin hayata ge\u00e7mesindeki en b\u00fcy\u00fck a\u015fama maliyetler. \u015eu an bu \u00e7al\u0131\u015fmalar laboratuvar ortam\u0131nda olduk\u00e7a y\u00fcksek maliyetlerle ula\u015ft\u0131\u011f\u0131m\u0131z sonu\u00e7lar. Dr. \u00c7a\u011favi de merkezde \u00f6zellikle kal\u0131t\u0131msal kalp hastal\u0131klar\u0131n\u0131n tedavi ve tan\u0131s\u0131na y\u00f6nelik \u00e7al\u0131\u015fmalar yapt\u0131klar\u0131n\u0131 aktararak, &#8220;Yapt\u0131\u011f\u0131m\u0131z kalp dokusu da bu \u00e7al\u0131\u015fmalar\u0131n bir par\u00e7as\u0131. \u00c7al\u0131\u015fmam\u0131z\u0131n temelinde \u00e7ok \u00f6zel bir k\u00f6k h\u00fccre grubu var. K\u00f6k h\u00fccreler, v\u00fccudumuzun embriyonik \u00f6ncesi hem de eri\u015fkin d\u00f6nemde yenilenmesi, onar\u0131m\u0131 ve geli\u015fimi i\u00e7in \u00e7ok \u00f6zel bir h\u00fccre grubudur.&#8221; dedi.<\/p>\n<p>\u00c7a\u011favi, \u00e7al\u0131\u015fmada \u00e7ok \u00f6nemli bir geli\u015fmeyi takip ettiklerini, uyar\u0131lm\u0131\u015f \u00e7ok \u00f6zellikli embriyonik k\u00f6k h\u00fccreye benzer bir h\u00fccre grubu \u00fcrettiklerini dile getirerek, \u015f\u00f6yle devam etti:<\/p>\n<p>&#8220;Bunlar\u0131, hastalar\u0131n ya da sa\u011fl\u0131kl\u0131 bireylerin herhangi bir v\u00fccut h\u00fccresinden alarak ki daha \u00e7ok kandan yap\u0131yoruz, kan h\u00fccrelerini izole ediyoruz ve ard\u0131ndan yeniden programlama dedi\u011fimiz bir teknikle bu h\u00fccreleri maruz b\u0131rakt\u0131\u011f\u0131m\u0131z kimyasallarla embriyonik k\u00f6k h\u00fccreye benzer h\u00fccreler elde ediyoruz. \u00d6zel teknik, s\u0131n\u0131rs\u0131z bir h\u00fccre kayna\u011f\u0131 olu\u015fturmam\u0131z\u0131 sa\u011fl\u0131yor. Ard\u0131ndan bu h\u00fccreleri, bu \u00e7al\u0131\u015fmay\u0131 kalp hastal\u0131klar\u0131 \u00fczerine yapt\u0131\u011f\u0131m\u0131z i\u00e7in kalp kas\u0131na ama ba\u015fka \u00e7al\u0131\u015fmalarda di\u011fer h\u00fccre tiplerine de farkl\u0131la\u015ft\u0131rmak m\u00fcmk\u00fcn. Yakla\u015f\u0131k bir ayl\u0131k bir s\u00fcre\u00e7 i\u00e7erisinde ald\u0131\u011f\u0131m\u0131z h\u00fccreler kalp kas\u0131 h\u00fccrelerine d\u00f6n\u00fc\u015f\u00fcyor. Tabii bu kalp kas\u0131 h\u00fccreleri embriyonik d\u00f6neme benzer yeni do\u011fan h\u00fccreler. Ard\u0131ndan da h\u00fccrelerin bir s\u00fcre daha k\u00fclt\u00fcre edilmesiyle kalp dokusunu kolayl\u0131kla g\u00f6r\u00fcyoruz. Bu bizim i\u00e7in son derece heyecan verici oluyor. \u00c7\u00fcnk\u00fc kas\u0131lmalar\u0131 ger\u00e7ek zamanl\u0131 olarak bazen mikroskopta, bazen buna bile gerek kalmadan g\u00f6rebiliyoruz.&#8221;<\/p>\n<p>Bu \u00e7al\u0131\u015fmayla h\u00fccrelerin molek\u00fcler olarak karakterize edildi\u011fini bildiren \u00c7a\u011favi, h\u00fccrelerin \u00fczerinde ila\u00e7 denemeleri i\u00e7in platform haz\u0131rland\u0131\u011f\u0131n\u0131 s\u00f6yledi.<\/p>\n<p>\u00c7a\u011favi, ayn\u0131 zamanda h\u00fccreler \u00fczerinde elektrofizyolojik \u00f6l\u00e7\u00fcmler yapt\u0131klar\u0131n\u0131 belirterek, &#8220;Bu h\u00fccreler bize, e\u011fer hasta bireylerden elde edersek hem s\u0131n\u0131rs\u0131z bir kaynak sa\u011fl\u0131yor hem de bu hastal\u0131klar\u0131n laboratuvar ortam\u0131nda ara\u015ft\u0131r\u0131lmas\u0131 i\u00e7in bir platform haz\u0131rl\u0131yor. Bu da bize hastal\u0131\u011f\u0131n laboratuvar ortam\u0131nda modellenebilmesini sa\u011fl\u0131yor.&#8221; ifadelerini kulland\u0131.<\/p>\n<p>Esra \u00c7a\u011favi, \u00e7al\u0131\u015fmayla herhangi bir molek\u00fcl\u00fcn kalp h\u00fccreleri \u00fczerinde olu\u015fturabilece\u011fi toksik etkileri saptayabildiklerini ya da yeni geli\u015ftirilen kalp hastal\u0131klar\u0131na y\u00f6nelik ila\u00e7lar\u0131n hem hasta hem de sa\u011fl\u0131kl\u0131 h\u00fccreler \u00fczerindeki etkilerini g\u00f6zlemleyebildiklerini s\u00f6yledi.<\/p>\n<p>Kalp hastalar\u0131ndan al\u0131nan h\u00fccrelerde hastal\u0131\u011fa neden olan mutasyonlar oldu\u011funu aktaran \u00c7a\u011favi, &#8220;Bunlar\u0131 yine genom m\u00fchendisli\u011fiyle de\u011fi\u015ftirebiliyoruz, d\u00fczeltebiliyoruz.
